Identification of Mycoplasma spp in bovine milk with an immunoperoxidase assay

ABSTRACT
The objective of this study was to implement an immunoperoxidase assay for the identification of Mycoplasma spp from bovine milk. Eighty samples of naturally infected milk, with different Mycoplasma spp, were analyzed; the immunofluorescence assay was used as a gold standard. The sensitivity of the immunoperoxidase assay was six colony-forming units, with no cross-reactions between the antisera, with 100% epidemiological sensitivity and specificity, and predictive values; there were no statistical differences (P › 0.05) between both tests. The reproducibility of the immunoperoxidase assay was high, obtaining identical results four times. Furthermore, it was carried out in less time than the immunofluorescence assay, requiring only a light microscope to read the results and fewer antisera and conjugates. In this study, the immunoperoxidase assay proved to have similar diagnostic value to the immunofluorescence assay, but was easier to perform and did not require sophisticated equipment.
