Reproduction of an experimental model for abdominal sepsis

ABSTRACT
Objective: To reproduce an abdominal sepsis model in the experimental animal.
Setting: Department of Surgery, School of Medicine, UNAM.
Design: Descriptive, comparative study.
Statistical analysis: We compared groups at 24, 48, and 96 hours, using Mann-Whitney U-test and Kruskall-Wallis.
Material and methods: We operated 18 Wistar rats, divided in 3 groups. Group I was subjected to laparotomy and wall closure. Groups II and III were subjected to laparotomy and the base of the cecum was ligated. Three punctures were made in the cecum with a 20-gauge needle. At 24 hours, all three groups were subjected to Laparotomy and samples from the abdominal cavity were taken for culture. The abdominal cavity of Group III was additionally washed with saline solution and closed. At 48 and 96 hours a new laparotomy was performed and samples were taken again from the abdominal cavity.
Results: Differences between Group I and Groups II and III were statistically significant. Comparison between the two last ones at 48 hours revealed no differences in the number of CFU. At 96 hours, differences were only found between the control Group (I) and Group II (
p 0.010). Between groups I and III no differences were found (
p 0.076); however, this could be due to a possible effect of the saline solution washing that made conditions similar between these two groups.
Conclusions: It was statistically demonstrated cecal ligation and puncture induces a significant increase in the number of CFU/ml, and that washing of the abdominal cavity with saline solution leads to a decrease in the number of CFU/ml.
