Utilidad de las interacciones farmacocinéticas/farmacodinámicas de analgésicos

RESUMEN
El presente trabajo constituye una revisión bibliográfica sobre las interacciones farmacocinéticas/farmacodinámicas determinadas con fármacos analgésicos-antiinflamatorios no esteroides en estudios preclínicos. Como introducción se revisan algunas definiciones elementales; después se comentan los modelos experimentales para evaluar el dolor, con especial énfasis en un modelo de dolor inducido por la disfunción de una articulación en ratas y finalmente las interacciones. De manera general, la farmacocinética estudia el curso temporal del fármaco dentro del organismo y la farmacodinamia los efectos farmacológicos que originan estos compuestos. El modelaje farmacocinético/farmacodinámico, aporta elementos que permiten establecer la combinación apropiada de analgésicos, tanto en tipo como en proporción, para un adecuado manejo del dolor. Básicamente, con estos estudios se logra asociar datos plasmáticos y terapéuticos mediante una ecuación matemática útil para la predicción. Por otra parte, en estudios preclínicos con modelos animales, la adición a ciertos analgésicos de compuestos adyuvantes como la cafeína, ha probado potenciar el efecto terapéutico de estos fármacos reduciendo la dosis administrada de los compuestos originales y por tanto, los efectos adversos que los analgésicos por sí solos presentan. Es importante difundir este conocimiento a los profesionales de la salud con el fin de ofrecer una mejor calidad de vida a los pacientes.
