Comparative study of two deproteinization methods for the evaluation of whole blood and plasma by magnetic resonance spectroscopy

ABSTRACT
Even though magnetic resonance spectroscopy is a very useful method in biological research, different issues have contributed to its minor diffusion in the clinical environment: i) high instrumental and experimental complexity, as well as, in the biochemical interpretation of measurements and ii) lack of automatized system providing reliable and accurate information. In this work, a number of metabolites from proton magnetic resonance spectrum extracts of whole blood and plasma previously prepared by acetonitrile and perchloric acid precipitation were obtained. Usefulness of both deproteinization methods, to be used in the future in a combined study using magnetic resonance and pattern recognition techniques, was assessed. Results demonstrated that both methods (perchloric acid and acetonitrile precipitation) produce good spectra including a lot of metabolites; however, acetonitrile is a more selective solvent providing an alternative method for spectra simplification while supplying enough information for the development of automatized classification methods via magnetic resonance information.
