Human leukocyte antigens class II and hepatitis C virus infection

ABSTRACT
Hepatitis C virus (HCV) persists in the majority of infected
individuals and is responsible for a wide spectrum of
chronic liver lesions ranging from minimal inflammation
to cirrhosis or hepatocellular carcinoma. Differences in
the immunogenetic backgrounds of infected patients might
in part account for the observed variation in the individual
courses of the disease. A genetically determined resistance
or susceptibility to chronic hepatitis C virus infection
make an important contribution to the course of liver
disease and may be linked to the Human Major Histocompatibility Complex (MHC). Several studies have aimed to identify human leukocyte antigens (HLA) class II alleles
associated with different outcomes of HCV infection. The
aim of this review is to make a differentiation between the
effects of some MHC class II alleles in the host response to
HCV infection. We found in the consulted literature alleles
like DRB1*11, DQB1*0301 and DRB1*0101 with a protective
role in the outcome, clearance of the HCV infection
and response to treatment and DQB1*02, DRB1*0701 alleles
with harmful effects in HCV infected patients. The
study of the role MHC class II alleles in the outcome and
clearance of HCV infection is important in understanding
the immune-virus specific response in the host and it will
improve the development of effective vaccines and antiviral
therapies.
