Evaluation of rivanol test for diagnosis of brucellosis in goats

ABSTRACT
The goal of this study was to evaluate the rivanol test for the confirmation of the diagnosis of brucellosis in goats. Sensitivity and specificity were also determined. The test was performed with a commercial antigen of
Brucella abortus. Dilutions used were 1:25, 1:50, 1:100, 1:200 and 1:400. For the sensitivity determination, sera from 64 infected goats with a positive isolation of
B.melitensis were used. Specificity was evaluated using 50 sera from brucellosis free goats. Specificity in vaccinated goats was determined using sera from 22, 3 to 4 month old goats vaccinated with Rev 1 (1 ´ 10
9 cfu/ml), and 59 adult goats vaccinated with a reduced dose of Rev 1 (1 ´ 10
5 cfu/ml). Sera were collected on days 7, 15, 30, 60, 90, 120, 150, 180 and 210 after vaccination. In the infected group, 54 animals were positive. Titrers varied from 1:25 to 1:400; and sensitivity was 83%. There were no positive reactors to the rivanol test in the Brucella free animals, resulting in 100% specificity. In the vaccinated group with the standard dose, all animals reacted from days 7 to 90; 27% of the animals reacted on days 120 and 150, and 9% of the animals from days 180 to 240 were positive. Sera from the goats vaccinated with the reduced dose reacted with a 100% on days 7 to 90; 35% on days 120 and 150, 21% on days 180, and 6% both on days 210 and 240. It is concluded that the rivanol test does not offer advantages to be used as a confirmatory test for brucellosis in goats.
