Influence of the vehicle in the oral acute toxicity of vainillin in Sprague Dawley rats

ABSTRACT
Introduction: vainillin has been proposed as therapeutic candidate in sickle-cell anaemia treatment, a disease for which effective treatment does not exist. In the
studies of oral acute toxicity, the election of the vehicle is a very important factor, mainly when the product is poorly soluble in water.
Objective: to evaluate the influence of dimethylsulfoxide and almond oil used as vehicles in the oral acute toxicity of vainillin in Sprague Dawley rats.
Methods: vainillin was administered by gavage at concentrations of 0,33-2 000 mg/kg: Rats were observed during 14 days for signs of toxicity.
Results: almond oil did not cause lethality, where as mortality was observed in the rats receiving 2 000 mg/kg of vanillin dissolved in dimethylsulfoxide. The macrocospic
analysis revealed that these animals presented autolysis and catarrhal enteritis while those receiving vanillin dissolved in almond oil showed steatorrhoeic hepatosis.
Conclusions: this investigation proved the importance of a good selection of the vehicle and how it can influence in the toxicological interpretation of results. Since the
preclinical studies are intended to guarantee the harmlessness and the maximum degree of safety of a product, it was concluded that in such circumstances, vainillin is
not completely innocuous when is administered by oral route to rodents. The observed effects depend on the proper selection of the vehicle to use in the preclinical studies.
