In vivo mutagenic evaluation of D-004

ABSTRACT
Introduction: D-004, a lipid extract from the fruits of royal palm tree (
Roystonea regia), has proved to be pharmacologically active in different experimental models of
prostatic hyperplasia.
Objective: to assess the
in vivo genotoxic effect of D-004 in orally administered male mice.
Methods: male mice were treated by oral gavege at doses of 500, 1000 and 2000 mg/kg of D-004 in micronucleus assay, chromosome aberrations test and comet assay. And with doses of 500, 1000 and 1500 mg/kg in the morphology assay of the spermatozoid head shape . Cyclophosphamide was used as positive control.

Results: Bone marrow micronucleus: D-004 did not change the frequency of micronucleated polychromatic erythrocytes , neither diminished the ratio between
polychromatic and normochromatic erythrocytes compared with the controls. Non significant differences were found in the spermatozoid concentrations, nor in the frequency of normal or abnormal spermatozoids among the groups.
Chromosomal
aberrations: D-004 did not induce chromosomal aberrations compared with negative controls.
Comet assay: No single induction of strand breaks or alkali-labile sites on DNA was observed as a consequence of the administration of the D-004.
Sperm morphology: There were no significant differences in the sperm concentration and frequency of normal and abnormal head shape of sperms among the different groups.
Conclusions: these
in vivo results indicate that D-004 does not show evidence of genotoxic activity both in germinal and somatic cells of rodents.
