Ghrelin: a regulatory hormone of food intake and body weigth

ABSTRACT
Body weight depends on the balance between energy intake and energy expenditure, when the intake is greater than expenditure obesity appears, a disease that endangers the lives of million of people worldwide. Food intake is regulated by a complex system that includes both central and peripheral factors. Ghrelin, a gastrointestinal peptidic hormone, is a potent regulator of food intake and body-weight. Subjects under fi xed feeding schedules display an increase of ghrelin secretion before mealtime, and decreases after food intake. Peripheral administration of ghrelin promotes hunger and food intake. In addition, plasma ghrelin levels are elevated in obese subjects, and become even more elevated after diets and surgeries aimed to lose weight. Based on studies describing the function and action of ghrelin, pharmaceutical companies have become interested on developing a ghrelin receptor antagonist that might help to treat obesity. Among the therapeutic strategies to control body weight and obesity, immunization against ghrelin seems to be one of the most promising. In this review we describe the physiology of ghrelin and studies of this hormone conducted in our laboratory, using the rabbit as a model. We also discuss the possible therapeutic importance of this hormone to control body weight in humans.
