Histamine and intercellular communication: 99 years of history

ABSTRACT
Since the description of the histamine actions on intestinal smooth muscle contraction and vasodilatation by Dale and Laidlaw (1910), numerous other effects have been added to the list of histamine-mediated actions. In the periphery, histamine participates in cardiac contraction force and beating rate, contraction of the smooth muscle in the respiratory and gastrointestinal tracts, acid gastric secretion and immune and allergic responses. In the central nervous system, histamine regulates several functions such as the sleeping-awaking cycle, motor behaviour, feeding and drinking, sexual behaviour, learning and memory. Histamine actions are mainly mediated by G protein-coupled (metabotropic) receptors, of which four of such receptors (H1-H4) have been identified. In this work we review the mechanisms involved in histamine synthesis and degradation, the pharmacological and functional characteristics of histamine metabotropic receptors, the main functions mediated by these receptors, and evidence for the involve ment of histamine in some pathological conditions.
