Influencia endocrina y paracrina sobre la adipogénesis

RESUMEN
Se revisó la literatura científica actual sobre los mecanismos de desarrollo del adipocito. Se cuantificaron los resultados de los estudios
in vitro e
in vivo (diferentes líneas celulares mesodérmicas con potencial adipogénico) bajo la influencia de las sustancias más representativas con actividad hormonal (insulina, factor de crecimiento similar a la insulina, glucocorticoides, triiodotironina y cAMP.) y parahormonal (GFs y TNFa) sobre los factores de trascripción celular (C/EBP, PPAR y ADD1/SREBP1) para inducir tanto la diferenciación del adipocito, como los cambios del patrón de reclutamiento/proliferación en la célula madura. El resultado de esta investigación mostró que existe un mayor número de agentes inductores para la diferenciación del adipocito (66.7%) que de inhibidores (33.3%), que PPARg2 fue el principal factor de trascripción involucrado en la adipogénesis y TNFa en inhibir este efecto. Con la interpretación lógica de estas interacciones moleculares, se pueden obtener las bases bioquímicas y fisiológicas necesarias para el diseño farmacológico de compuestos antiobesidad que puedan actuar sobre estructuras celulares específicas relacionadas con la adipogénesis; además, el clínico puede incrementar sus conocimientos, tanto de la fisiopatología de la obesidad, como de la farmacodinámica de los nuevos medicamentos que se utilizan para tratar el exceso de tejido adiposo.
