Role of CYP2E1 genetic polymorphism in the risk of acute pesticide intoxication

ABSTRACT
Xenobiotic metabolism is dramatically influenced by the presence of allelic variants in the drug metabolizing enzymes. Several studies have indicated the presence of different expression levels of CYP 2E1, an enzyme involved in the metabolism of pesticides, resulting in a differential susceptibility to the chronic exposure to these compounds. Taking into consideration the board use of these compounds in agriculture, the aim of the present study was to establish the association of the genetic polymorphism of CYP2E1 with the intoxication symptoms in a population of sugar cane fumigators. The frequency of CYP2E1
RsaI/PstI and
DraI polymorphisms was determined by PCR-RFLP methods in DNA isolated from blood samples. In parallel, a questionnaire was prepared in order to detect clinic information concerning intoxication symptoms in the fumigators. Our results indicate that the mutated allele for
DraI is associated with a 4 fold- increase in susceptibility to pesticide exposure. Genetic polymorphism studies are imperative in exposed population in order to prevent or reduce occupational risks.
