Neurobiochemistry and pharmacology of the atypical antipsychotics

ABSTRACT
The knowledge about the existence of different neurotransmitters other than dopamine involved in schizophrenia, like serotonina and excitatory amino acids, has been relevant for the development of new molecules without extrapyramidal effects and other side effects. Such new antipsychotic drugs have been named for that reason atypical.
Also, it has been very important in that field to understand that the occupancy of less than 70% of dopamine D2 receptors at the striatum, is one of the pharmacological profile of the new atypical medications, along with a higher antagonism on serotonergic 5-HT2A receptors. New drugs as olanzapine, ziprasidone, aripiprazol, risperdone and quetiapine are described in the present review. Also the description of the neurotransmitter systems as well as some aspects of atypical antipsychotics’ pharmacodinamics are mentioned, too.
