Effects of deoxyribonuclease I on murine melanoma B16-F10 cells.

ABSTRACT
Introduction: Importance of the nucleases in the regulation of cellular growth (through the apoptosis) makes necessary to investigate their utility to control tumoral growth.
Objective: We evaluated the effect that deoxyribonuclease (DNase) had on cells of the melanoma murine B16-F10.
Material and methods: the cells were incubated at different times and concentrations of the DNase and then their viability, growth and damage to the DNA were evaluated.
Results: Showed that their viability and mitochondrial activity measured by means of the exclusion technique with trypan blue or with MTT didn’t show any significant change, however, the capacity of cellular growth (total number of cells) was modified in a dependent dose way. With regard to the damage to his DNA our results indicate an effect at the 24 hours of the nuclease incubation with the cells, bands of 400 and 200 pb were found in the agarose gel.
Conclusion: the DNasa used in this work didn’t have any significant effect on the viability, but did on the cellular growth, as well as on the structure of the DNA due to an uncertain mechanism.
