State of the art determination of uric acid, evaluated front the HPLC method

ABSTRACT
A reversed-phase HPLC method was used to determine the content of uric acid in eighteen proficiency samples based on equine serum (100%), that were in turn assayed by the Clinical Laboratories participating in the monthly Introducción surveys conducted by the Cuban External Quality Control National Program during the 1993- and 1994-year campaigns. The reported results were then distributed according with the chemical method locally used in the measurement of uric acid, and compared with the HPLC-value. The interval for the allowable deviation from the HPLC value included the median together with 50% of the results obtained locally with the phosphotungstic acid reaction. The use of this colorimetric method was associated with the highest proportion of outlying values. The move towards an automatized/mechanized method brought about a reduction in the frequency of outliers, and a marginal increase in the number of satisfactory results. The manual version of the uricase reaction was characterized by a deviated behaviour from the HPLC-value. The results presented in this article demonstrate the advantages of the inception of HPLC applications for Clinical Chemistry analytes in an External Quality Control Program.
