Effects of D-002 on the hepatotoxicity induced by paracetamol in rats

ABSTRACT
Introduction: Paracetamol, though widely used in clinical practice as antipyretic and analgesic produces hepatotoxicity as side effect. That is why, it is a concern to find treatments to prevent such effects. D-002, a mixture of aliphatic alcohols of high molecular weight obtained from sugar cane wax, has gastroprotective and antioxidant effects, and has shown to be preventive of CCl
4-induced toxicity in experimental models.
Objective: to evaluate the effects of D-002 in the Paracetamol-induced hepatotoxicity in rats.
Methods: animals were randomized into 6 groups: a negative control vehicle and five groups with Paracetamol: a positive control and four treated with D-002 (5, 25, 50 and 100 mg/kg). The treatments were administered for 7 days, and once the last dose was applied, the damage was induced half an hour later. The animals were sacrificed 24 hours after damage induction. Histopathological analysis of liver tissue samples was performed; the enzymatic activity of transaminases was quantified too.
Results: all positive controls and any of the negative ones showed the pattern that characterizes hepatotoxic damage for this agent. D-002 (5-100 mg/kg) reduced the histological damage by increasing the average of normal hepatocytes compared to the positive control and to the average of damaged cells. In addition, transaminase activities increased by Paracetamol were also decreased by D-002.
Conclusions: D-002 was effective to protect against Paracetamol-induced hepatotoxicity in rats.
