Effect of periodontitis in postpartum women in relation to the DNA damage, as well as its potential teratogen in the preterm labor newborn

ABSTRACT
Background: Micronucleus assay detects DNA damage. Periodontitis is a chronic degenerative disease of the oral cavity and the generation of free radicals is an important consideration in this disease that could result in an increased frequency of DNA damage. There is evidence in the literature suggesting an association between periodontal diseases in pregnant women and giving birth to premature infants. Also we have investigated whether pathologies that are associated with the formation of free radicals lead to an increase the frequency of micronucleus in preterm newborns during pregnancy. Objective: Evaluate DNA damage increased by means of micronuclei assay in buccal mucosa cells from puerperal women with periodontitis and to determinate the teratogenic potential in the preterm newborns to mothers with periodontitis by means of counting micronucleus in peripheral blood. Material and methods: Buccal mucosa cells from puerperal woman with periodontitis and without periodontitis and peripheral blood from preterm newborns were collected and spread directly on two separate, pre-cleaned and pre-coded slides. The smears were air-dried, fixed, stained and analyzed by microscopy. DNA damage was determined by counting micronucleus and nuclear abnormalities in exfoliated cells, and in peripheral blood the number of micronucleated erythrocytes, micronucleated polychromatic erythrocytes and the number of PCE. Conclusion: The increase in micronucleus and nuclear abnormalities frequency in buccal mucosa cells of puerperal women with periodontitis and the increased in micronucleated polychromatic erythrocytes number in preterm newborns to mothers with periodontitis indicates that the periodontitis per se increases DNA damage and its potential teratogenic effect on preterm newborns to mothers with periodontitis.REFERENCES
