Comparison between «in situ» sciatic nerve grafts and vein grafts with platelet-rich plasma. Experimental study in rabbits

ABSTRACT
Nerve grafts are a very useful weapon in reconstructive surgery of extremities. Joining two ends through a nerve interposition prevents excessive tension on the edges, reduces fibrosis and improves nerve regeneration. The major drawback of a nerve graft is the loss of feeling of the donor area. We carried out a pilot study phase I with eight male rabbits, strain Rex, weighing between 900 and 1,200 g each. We divided the population into two groups so that each included a rabbit leg. Group A was the control group with a sciatic nerve graft, and group B, the control group with a venous graft stuffed with platelet-rich plasma. After eight weeks we sacrificed the rabbits and analyzed the samples with hematoxylin-eosin and
Luxol fast blue tinctures. Seven of eight samples of group A and group B showed the presence of proximal axonal neuroma growth; however, the proximal neuroma in group B was smaller. All samples displayed demyelination observed with
Luxol fast blue staining. We concluded that in some cases the nerve grafts can be replaced by vein grafts with platelet-rich plasma to reduce morbidity of the donor area.
