Role of SRY Transcription Factor in the Expression of Wnt4

ABSTRACT
Background: Wnt4 gene is an autosomal gene that codifies for a protein that plays a role in the gonadal development. The promoter sequence of this gene contains a consensus sequence that potentially may be recognized by the SRY protein (testicular determining factor), and therefore regulate Wnt4 expression.Objective: To analyze the involvement of the SRY transcription factor on the expression of the promoter of Wnt4.
Material and method: We performed the SRY-TARGET construction, which was transfected into HEK293 cells (Human Embrionic Kidney 293). The presence of the protein in the cells was verified by Western blot. Results: We did not find a significantly changes in the mRNA level of the Wnt4 gene in the presence of the SRY protein.
Conclusions: To our knowledge, this is the first study focused on analysis of the expression regulation of Wnt4 gene in the context of the gonadal development. Since no effect was observed on the expression of Wnt4 gene in the presence of SRY protein, one possibility is that the consensus region of the gene is a target for other proteins such as Sox9 R-spondin1, which consequently may participate in the gonadal differentiation.
