ABSTRACT
Allelic association studies in schizophrenia have focused on different chromosomal regions that contain genes coding for dopamine receptors. The polymorphisms of the dopamine receptor D4 (DRD4) have been widely associated with psychiatric symptoms. There are some reports of association between the polymorphism 48-bp-VNTR in exon III and schizophrenia, although this relationship remains controversial. Most studies have used patients comparing to controls or trios; however, there have been no studies on this polymorphism using families with multiple affected subjects. In this study we analyzed the VNTR polymorphism of the DRD4 exon III in 60 families with at least one pair of affected siblings. Data was analyzed using SIBPAL (S.A.G.E. software). The average estimate of shared alleles was 0.54 (p=0.014) and the estimated proportion of siblings with one or two shared alleles was statistically significant 0.499 and 0.208 (p=0.01, p=0.004). These results suggest that the 48-bp-VTNR-DRD4 polymorphism may be associated with schizophrenia in Mexico City families. The association reported in this work could be positive due to the particular population mix in Mexico City.
