Effect of vascular endothelial growth factor on tracheal healing (tracheoplasty or self-transplantation) in rats

ABSTRACT
Background: Tracheal transplants longer than 6 cm are prone to poor healing due to ischemia. The vascular endothelial growth factor (VEGF) induces angiogenesis and favors tissue healing.
Objective: To evaluate the macroscopic, microscopic and immunohistochemical changes induced in trachea healing after tracheoplasty or autotransplantation treated with topical application of VEGF.
Material and Methods: Forty rats were operated on: group I (n = 10), tracheoplasty treated with saline solution; group II (n = 10), tracheoplasty treated with VEG F; group III (n = 10), autotransplantation treated with saline solution and group IV (n = 10), autotransplantation treated with VEG F. The animals were clinically evaluated during 4 weeks and at the end of the study the anastomosis was macroscopically, microscopically, morphometrically and immunohistochemically studied.
Results: Macroscopically, all the anastomosis healed. Nevertheless, 40% and 10% of groups II and IV, animals presented healing abnormalities. Groups II, III and IV morphometrics showed tracheal stenosis (p ‹ 0.05 Andeva, Tukey). The immunohistochemical study revealed a higher VEGF expression in group IV (p ‹ 0.01, Andeva, Tukey, Kruskal-Wallis).
Conclusion: The topical application of VEG F in tracheal autotransplants promotes angiogenesis and favors tracheal healing, but promotes scarring and tracheal stenosis.
