Motility and acrosomal damage of buck semen frozen in 0.25 ml and 0.5 ml straws and thawed at two different temperature rates

ABSTRACT
Sixteen ejaculates from 4 bucks were collected with an artificial vagina in order to evaluate volume, sperm concentration, progressive motility and acrosomal morphology. Split ejaculates were diluted and packaged in 0.25 and 0.5 ml straws, each containing 100 million motile sperms. Straws were frozen in liquid nitrogen vapour and thawed at 37°C/15 sec or 55°C/8 sec. Progressive motility, percentage of motility recovered and acrosomal damage was determined in thawed samples. Progressive motility after thawing was 62.8% in the 0.5 ml straws (37°C/15 sec) and 58.7% (55°C/8 sec); 59% in 0.25 ml staws (37°C/15 sec) and 60% (55°C/8 sec) ; (P › 0.05). Acrosomal damage was higher (12.3%) in 0.5 ml straws thawed at 55°C/8 sec, than in the other groups (P ‹ 0.05). Thawing at 37°C/12 sec is simpler and implies less risk. Progressive motility and acrosomal damage of the thawed semen found in this study proved to be similar to the standards established for artificial insemination in this species.
