Evaluation of an ultramicroELISA for the detection of infection by the respiratory syncytial virus.

ABSTRACT
Objective. The present study was aimed at evaluating antibody ultramicroELISA for rapid detection of IgG antibodies to Respiratory Syncytial virus in single and pair sera.
Materials and methods. 1567 serum samples were evaluated by both complement fixation (FC) and ultramicroELISA (UME) techniques: 619 single serum samples from children under 15 years and 474 pair serum samples from the national surveillance seroepidemiological program for acute Respiratory Diseases. In the double antibody UME technique, a specific monoclonal antibody against the F protein of RSV was used.
Results.When comparing the ultramicroELISA with the FC technique, values obtained for single serum samples were: sensitivity was 98.9 %, specificity 56.05% and agreement 63.9 %. And for the pair serum samples sensitivity was 92.7 %, while specificity and agreement increased in a 79.3 % and a 80.1 % respectively.
Conclusions. The use of this monoclonal antibody allowed the inclusion of rough antigen preparation in stead of purified fractions, leading to a remarkable decrease in the reactivity obtained with antigen control. These results back up the use of this method as a complementary tool in seroepidemiological studies carried out on a large scale and for diagnostic purposes.
