Detection of mycobacterium in sputum specimens, by amplification of the rDNA16S marker with the polymerase chain reaction

ABSTRACT
Tuberculosis dynamic in Mexico and the world is forcing to readdress the procedures against this disease. In this sense the molecular diagnosis by the acid nucleic amplification from Mycobacteria by the polymerase chain reaction, is been pointing as a technique with certain success in countries with medium and high prevalence of the disease. So the main goal of this work is to evaluate utility of the diagnostic of tuberculosis by the PCR technique, amplifying the rDNA16s marker, from sputum samples from patients with suspicious of tuberculosis. The results show that from 107 clinical samples analyzed, 36 were positive to PCR and 37 were acid fast bacilli smear, the sensibility and specificity of the procedure were 80 and 90% respectively and the positive and negative predictive value was 80 and 88%, the average time for the development of the procedure was 16 hrs. Despite that this is a preliminary study, our results let us to claim the utility of this fast and confidence diagnostic procedure, as an important tool against TB.
