Evaluation of the modified card test for diagnosis to goat brucellosis

ABSTRACT
Sensitivity and specificity of two different concentrations of the card test antigen for the diagnosis of Brucellosis in goats was evaluated. Antigens used were the oficial antigen produced by a national laboratory in Mexico ("Productora Nacional de Biológicos Veterinarios") with a cellular concentration of 8%, and an experimental antigen with a cellular concentration of 3%. Both antigens were made using
Brucella abortus strain 1119-3 in a lactate buffer with a pH of 3.6. Sensitivity for the two antigens was determined using 53 sera from
Brucella melitensis culture positive goats. Sera from 100 goats, negative to brucellosis and collected in a brucellosis free area were used for specificity determination. Sensitivity for the 8% cellular concentration was 79%. When the concentration was of 3%, sensitivity was 98%. Specificity for both antigens was 100%. It is concluded that the diagnosis of brucellosis in an 8% cellular concentration is not adequate due to its low sensitivity. The use of a 3% cellular concentration in the card test antigen for goats is recommended.
