A comparative morphological study of dopaminergic grafts among different basal ganglia

ABSTRACT
Fetal mesencephalic graft into dopaminergic denervated structures is one of the restorative strategies used in the treatment of Parkinson’s disease. In the present study, we compared the morphological features of dopaminergic grafts into the striatum, the subthalamic nucleus and substantia nigra pars reticulata. Material sand methods: wistar rats with unilateral 6-hydroxydopamine lesions of the substantia nigra pars compacta were divided into three groups and received injections of 300 000 embryonic rat ventral mesencephalic cells in the striatum, substantia nigra pars reticulata, orsubthalamic nucleus. Graft survival was assessed using tyrosine hydroxylase immunohistochemistry, 4 month safter transplantation. Results. Intranigrally and intrasubthalamically grafted cells, unlike intrastriatally graftedones, spread toward several surrounding areas. The dispersion of dopaminergic neurons was greater in the ones implanted into the subthalamic nucleus. In the three cases, TH-immunoreactive cells tended to clusterin the periphery of the grafts, with fewer cells observed in its center. A network of TH-positive fibers was observed within the graft itself and also extended into the host in all directions. Dopaminergic neurons grafted in the striatum showed more developed neuriticprocesses and larger cell bodies. Besides, we have found that the graft volume and TH-positive cell numberwere greater in the intra striatal grafts than in the intrasubthalamic or intranigral grafts.
