Current antifungals and future therapeutic targets: A review article

ABSTRACT
Invasive fungal infections have become a threat to human
health due to the high morbidity and mortality rates they are
responsible for, mainly in immunocompromised patients. The
available armamentarium for the treatment of these infections
is restricted to a minimum amount of antifungals from the
polyenes, the azoles, the echinocandins, the allylamines and the
pyrimidines classes, with high toxicity, drug-to-drug interactions,
and distribution problems as a result of the lack of administration
routes. Given that there is an urgent need to develop new
antifungal drugs, multiple substances are currently in advanced
stages of research, while other molecules continue to be investigated
for this end, including the orotomides, the terpenoids,
the glycosyl-phosphatidylinositol inhibitors, the tetrazoles, the
arylamidines, and the siderophobes, among others. This review
has the purpose of informing about the new classes of antifungals
and the suggestive pathways for possible therapeutic
targets.
