Efectos del pretratamiento con propofol sobre la endotelina en daño pulmonar agudo inducido por ácido oleico

RESUMEN
Introducción. El daño pulmonar agudo (ALI) es una patología
caracterizada por lesión de la membrana alvéolo-capilar
que origina un proceso inflamatorio tanto local como sistémico.
La endotelina (ET) es una proteína que actúa como modulador
inmune, aunque un incremento en su síntesis puede
contribuir al daño pulmonar.
Objetivo. Evaluar si el propofol
atenúa las alteraciones hemodinámicas, gasométricas,
microscópicas, inmuno-histoquímicas y de concentración
plasmática de ET-1 en un modelo experimental de daño pulmonar
agudo inducido por ácido oleico (OA).
Material y métodos.
Los perros mestizos incluidos en el estudio se dividieron
en tres grupos de estudio: grupo I testigo, grupo II ALI producido
con OA sin tratamiento y grupo III pretratamiento con
propofol y ALI inducido con OA.
Resultados. Todos los animales
sobrevivieron al tiempo de estudio. Los hallazgos hemodinámicos,
gasométricos y gravimétricos revelaron que los
grupos II y III desarrollaron ALI. Sin embargo, el grupo III
mostró menor infiltrado inflamatorio y expresión de ET-1 en
tejido pulmonar.
Conclusiones. El pretratamiento con propofol
en un modelo canino de ALI provocado por OA indica
que el fármaco presenta acción antiinflamatoria, con un potencial
papel terapéutico contra la progresión de la inflamación
y daño pulmonar.
