Siendo el agua la fuente esencial para las funciones vitales de cualquier organismo, no es raro pensar que la falta o solidificación de ésta cause incompatibilidad para la ida. Sin embargo, la permanencia de una célula en estado de congelación se contrapone a lo anteriormente dicho, ya que con el método de congelamiento se puede preservar una célula a temperaturas extremadamente bajas, permitiendo que el metabolismo se reduzca "absolutamente", sin que pierda su potencial vital. A temperaturas de -196°C no hay reacciones bioquímicas ni energía térmica dentro de la célula, aún más, no hay evidencia de que puedan haber cambios de índole genético. No obstante, dicha estabilidad solamente puede ser mantenida a temperaturas por debajo de los 130°C, ya que, a temperaturas mayores, puede haber agua no congelada intracelularmente la cual permite funciones metabólicas, causando degradación de la célula.
