Modificaciones calcio dependiente de radicales libres en sinaptosomas de corteza frontal de ratón por la ingesta crónica de alcohol

RESUMEN
El presente trabajo analiza los efectos del consumo crónico de etanol sobre diferentes parámetros que inducen estrés oxidativo y sobre el comportamiento bioenergético de los sinaptosomas; ya que es bien conocido que el consumo excesivo o crónico de alcohol se asocia a diferentes procesos degenerativos e inflamatorios en el sistema nervioso central (SNC), además de producir un efecto depresor sobre diferentes centros nerviosos inhibidores. Esto puede ser debido al aumento de fluidez en las membranas y/o a cambios en la función de las proteínas que se insertan en ellas.
Material y métodos: se determinán la formación de radicales libres, el nivel de peroxidación lipídica y el grado de oxidación de proteínas como índices del estrés oxidativo, y la actividad mitocondrial como índice del comportamiento bioenergético en sinaptosomas de corteza frontal de ratones controles y ratones que tomaron etanol, en situaciones basales y despolarizantes (K
+ 25 mM), en presencia o ausencia de calcio en el medio.
Resultados: se observó un incremento en la formación de radicales libres tras la ingesta de etanol en presencia de calcio en el medio, que no se relacionó con incre-mentos en los niveles de peroxidación lipídica ni con oxidación protéica. Tampoco se encontró relación entre la actividad mitocondrial y la formación de radicales libres, como había sido observado en estudios previos, realizados in vitro.
Conclusión. el presente estudio demuestra que la ingesta in vivo de alcohol provoca alteración en los que el Ca parece ser un factor determinante.
