ABSTRACT
O-linked N-acetyl-β-D-glucosamine glycosylation or O-GlcNacylation (O-GlcNAc) is a
post-translational modification consisting on the addition of N-acetyl-β-D-glucosamine
(GlcNAc) to serine and threonine residues in proteins. Its effects on cells are dual,
however, when this modification is expressed chronically, as happens in response to
hyperglycemia in patients with diabetes mellitus, its effects tend to be adverse, one
of its targets being endothelial cells. Because endothelial dysfunction is caused by
both O-GlcNAc and oxidative stress, it has been hypothesized that both processes
are interrelated, and that O-GlcNAc is probably responsible for the adverse effects
of oxidative stress in endothelium. Nrf2 could be a key piece in the O-GlcNAcoxidative
stress relationship in endothelial dysfunction, since, in the first place,
this transcription factor controls the basal expression and induction of antioxidant
enzymes, and recently, it has been demonstrated that its activity could be regulated
by O-GlcNAc. Therefore, this review describes the most recent investigations on
the role of O-GlcNAc in the endothelium and proposes the inactivation of Nrf2 as
an important factor in the development of endothelial dysfunction.
