Immunological studies in the donor/recipient pair for Hematopoietic progenitor cells transplantation

ABSTRACT
Pretransplantation results of immunological studies were analyzed (HLA donor and
related recipient typing and measurement of anti HLA class I and class II receptors)
for hematopoietic progenitor cells transplantation in the period from January to
December, 2013 at the Center for Cell Engineering and Transplantation (CICEL) of the
Institute of Hematology and Immunology (IHI). Molecular typing of HLA loci A, B, DR
and DQ was performed withOlerup SSP HLA SSP AB comby tray DR DQ low resolution
kit. To determine the presence of antibodies to HLA immunoenzimatic technique
(ELISA), LIFECODES QuikScreen LIFECODES Kit B-Screen anti-HLA class I and II was
used. Acute leukemias were the most frequent diagnoses Of all potential donors
45,6% were haploidentical with the receptor; totally incompatible 21%, and 33,3%
were identical. Regarding the HLA polymorphism in the population studied, for class I,
locus A: antigens A*02 and A* 24 were the most common in all individuals. For locus
B, antigens B*35 and B* 44 prevailed in white persons; and B *35 and B *18 in
nonwhites. For class II DRB1 locus, DRB1*03 and DRB1*04 antigens were common in
white; in nonwhite individuals DRB1*15 accompanied DRB1*03; for locus DQB1
antigens DQB1*03 and DQB1*05 were frequent for whites, and DQB1*06 for nonwhites.
The major incompatibilities occurred in loci B and DRB1. The population tested
showed high presence of anti-HLA antibodies, 44% for class I and 72% for class II.
