Thalidomide: From darkness to brightness

ABSTRACT
After the well known thalidomide tragedy that caused severe
birth defects in the late fifties and early sixties, this drug has
been successfully reborn. Currently, physicians recognize its
usefulness for treating several diseases, despite the fact that the
Food and Drug Administration (FDA) has only approved its use
for erythema nodosum leprosum and multiple myeloma.
Thalidomide has several mechanisms of action, and among
them, its anti-inflammatory, immunomodulator and anti-angiogenic
effects have been extremely useful in dermatology.
Therapeutic results have been promising in most of the cases.
The best results have been reported in actinic prurigo, erythema
nodosum leprosum, aftous stomatitis, Behçet’s disease, erythematous
lupus, and nodular prurigo. Nonetheless, its safety is
still a source of great concern and controversy in the medical
literature.
