Determination of Levels of Lymphocytes Subpopulations in Healthy Adult Donors at CMN 20 de Noviembre, ISSSTE

ABSTRACT
Background: Clinical diagnosis has undergone several modifications
due manly to the advances generated by the new methods of
cellular analysis. Flow cytometry identifies different lymphocytes
subsets for the diagnostic and evaluation of a great variety of
illnesses. For that, it is necessary to determine the variation and
reference values in healthy subjects.
Objective: To know reference values for CD3, CD4, CD8 and
CD4/CD8 analyzing values of lymphocytes subsets in healthy
donors by flow cytometry.
Material and method: A descriptive, cross-sectional and comparative
study was done with 51 healthy adults (30 male and 21
female) of 18 to 57 years old from Central Blood Bank of National
Medical Center 20 de Noviembre, Mexico City, were selected. The
determination of subsets of lymphocytes T CD4+ and T CD8+ was
carried out by lysis non laundry with flow cytometry FACSCalibur
of Becton Dickinson.
Results: Results and reference intervals obtained for the total
group were: for CD3 a media of 194, interval of 852-3,264, with
p (‹1) 0.78; for CD4, a media of 1,145, interval of 580-1,944 and
p (‹1) 0.40; for CD8, a media of 771, interval of 264-1,533 and
p (‹ 0.05) 0.10; for the relationship CD4/CD8, a media of 1.67,
interval of 0.59-3.1 and
p (‹1) 0.063.
Conclusions: Data obtained were correlated with previous values
used in hystocompatibility laboratory, and significant differences
were observed. It is necessary to perform more extensive research
in order to compare results.
