Polymorphisms of superoxid dismutase in carotid aterosclerosis and stroke

ABSTRACT
Background: Functional polymorphisms in the genes encoding the superoxide dismutase (SOD), a powerful antioxidant enzyme, may play an important role in the pathogenesis of stroke and atherosclerosis disease. Methods: The allele frequencies of two polymorphisms of SOD genes (Mn and CuZn) were investigated in 140 patients with ischemic stroke and compared with 140 controls. We conducted a descriptive analysis of the demographic variables, through analysis of univariate association using χ2 test, the Mann-Whitney U test and Student’s T test, respectively. The genetic variables were included in a Cox regression analysis model paired by age, gender and etiology, with p < 0.05 level significance. Results: Of the 140 patients with stroke, 39% were women and 61% were men. The average of age was of 63, 1 ± 1, 37 years old; 46% carotid intima-media thickness (IMT) › 1 mm, 72% carotid plaque (IMT › 1.5 mm) and 33% carotid stenosis (› 30%). In stroke patients 98.5% were homocigotes and 1.5% heterocigotes for CuZnSOD; and for MnSOD polymorphism 50% were homocigotes for alanine, 6.4% homocigotes for valine and 43.5% heterocigotes alanine/valine. No differences with controls were founded (p › 0.05). We found there was no association between the polymorphisms of the MnSOD and the CuZnSOD and the degree of carotid atherosclerosis or stroke. Conclusions: Was not associated between the polymorphisms of the MnSOD and the CuZnSOD with the presence of stroke or with carotid atherosclerosis. Further studies are required in different populations in order to establish a conclusion.REFERENCES
