Relation betwen acrosomal reaction, sperm motility, acridine orange and sperm penetration essay with ascorbic acid

ABSTRACT
Objective: To determine the use of ascorbic acid (AA) as an antioxidant treatment protects DNA spermatozoa integrity; spermal motility and acrosomal reaction.
Design: Through 48 hr incubation control (group 1), 300 µM AA (group 2), 50% follicular fluid egg donor (group 3), 50% follicular fluid IVF failed patient (group 4) spermatozoa of 52 patients under infertility study were analyzed and compared.
Results: Motile sperm survival 67 ± 3% at 300 µM AA, spermatozoa supplementation with AA showed a significant positive relationship of motility after 24 hr (p ‹ 0.005) with AA treatment. The percentage of acrosomal reaction after 24 h incubation was 35.35 ± 2.2 and 50.1 ± 3.1 after 48 h (p ‹ 0.001) there were insignificant with the treatment. After 24 hr treatment the percentage of DNA integrity was 82.72 ± 1.78 and after 48 hrs 92.66 ± 1.76 (p ‹ 0.001) there were no significant difference with the treatment. After 24 hr in 60% of cases the number of ova penetrated and number of penetrations were higher in AA group than control however there was no significant difference with the treatment.
Conclusions: After 24 h incubation ascorbic acid increases the motility of spermatozoa and increases the acrosomal reaction. It is possible to use the AA as inductor of acrosomal reaction and as protector of DNA integrity. The difference at the SPA was statistically significant in AA group; however, the effect in the number of ova penetrated and number of penetrations are present and we consider that if we increase the number of patients the difference will be significant.
