Increment in p53 protein in leukocytes from rats with sodium fluoride toxication

ABSTRACT
Introduction. The subjects intoxicated with sodium fluoride (NaF) they present consecutive infections in lung and digestive system. It is hypothetized that this latter substance can have some effect upon the immunologic system. The objective is to determined the effect of the NaF in the leukocytes from rats intoxicated with NaF using p53 protein determination used like sign of damage.
Methods: Male rats was dealt with two concentrations of NaF (1 and 50 ppm) for eight weeks. As control, rats without NaF was used. Animals was sacrificed and blood obtained for leukocyte isolation. Leukocyte was analyzed for viability and detection of p53 protein by Western-Blot method.
Results: Along the time of treatment, the leukocytes from rats intoxicated with 50 ppm of NaF present a lower viability and an increase in p53 presence has compared with both the control group and rats intoxicated with 1 ppm of NaF.
Conclusions: Our results shows that the intoxication with NaF has can induced an increase in p53 protein. These latter protein is an important indicator of DNA damage. These can be interpreted like a genotoxic damage to the leukocytes.
