Isolation and identification of Actinobacillus pleuropneumoniae in pigs lungs with chronic pleuropneumonia at the slaughterhouse of Merida, Yucatan, Mexico

ABSTRACT
Introduction. The objective of this study was to standarize the direct isolation test technique and to develop the quick aglutination test for the isolation and identification of
Actinobacillus pleuropneumoniae in lungs from pigs with cronic pleuropneumonia. The present study was carried out at the slaugtherhouse of Merida, Yucatan, Mexico.
Materials and Methods. From July 1996 to February 1997, two hundred and fifty pig lungs with pleuropneumonia lesions in the caudal lobes were collected from the slaughterhouse of Merida, Yucatan, Mexico. Samples were cultured in blood agar with a nurse strain of
Staphylococcus aureus, they were incubated at 37°C for 24 hr. in aerobiosic conditions. The colonies that showed satelitism beta haemolysis and brightness were considered for further analysis. For the
A pleuropneumoniae identification the biochemical test of urea, glucose, lactose, mannitol, dulcitol, CAMP y NAD (micromethods) were used. The slide agglutination test was used for serotyping of
A pleuropneumoniae.
Results. In 129 (51.6%) lungs
A pleuropneumoniae was isolated. In 113 (87.6%) samples
A pleuropneumoniae was isolated in pure culture, in 16 (12.4%) other bacteria were present (15 samples with
P multocidae and 1 with
A pyogenes). The
A pleuropneumoniae identified serotypes were: serotype 1 in 88 (68.2%) strains, serotype 7 in 29 (22.5%), serotypes 5 in 1 (0.8%) strain and crossreaction between 1 and 7 serotypes was observed in 11 (8.5%) strains.
Discusion. The direct isolation technique for the culture of
A pleuropneumoniae in pig lungs with chronic pleuropneumonia can be used for the diagnosis of this disease. Sample selection is one of the most important factors in order to isolate
A pleuropneumoniae by the direct isolation technique.
