Indometacina protege de los efectos del ozono sobre el sueño

ABSTRACT
Ozone (O
3) exposure cause extrapulmonary effects, whose mechanisms are unclear, since O
3 do not pass beyond the pulmonary tract. An involvement of prostaglandins (PG) and nitric oxide (NO), two molecules produced by reactions between O
3 and the surfactant factor, has been proposed. In order to assay that hypothesis, we administrated indometacin (IND, 4 mg/kg), a PG and NO inhibitor, to eight male wistar rats exposed to 1 ppm of O
3. Sleep was the extrapulmonary toxicity model employed; so, animals were implanted in order to record polygraphically sleep states. Sleep recordings were taken during 24 hours in every of the experimental conditions: 1. clean-air exposure; 2. IND + clean air; 3. 1 ppm of O
3 exposure, and 4. IND + O
3 exposure. It was found an increase of slow waves sleep and a concomitant decrease of paradoxical sleep when rats were exposed to O
3. IND showed a protective effect mainly on paradoxical sleep. Regarding its role in sleep maintenance, results suggest that NO production and accumulation could be primarily involved in the extrapulmonary toxicity induced by O
3 exposure
