Apolymorphonuclear leukocyte depletion model in 5-fluorouracil treatment chickens

ABSTRACT
Fifty one-day-old broiler chicks were used in this study in a polymorphonuclear leukocyte (PMN) depletion model with 5-fluorouracil treatment chicken. Forty-five animals were randomly assigned into 3 groups of 15 birds each, and 5 chicks were used for bacteriologic monitoring. On day 17, the control Group (1) was intravenously inoculated with sterile phosphate-buffered solution (2 ml PBS on average) and pH 7.4, and Groups 2 and 3 were inoculated with a single dose of 200 and 300 mg/kg body weight 5-fluorouracil(5-FU) to each bird, respectively. On days 1,3,5,6,7,8,9,10,12 and 15 postinoculation (PI), 10 chicks from each group were bled for counts of total- and differential white blood cells and determination of polymorphonuclear leukocytes/total leukocytes (PMN/TL) ratio per day. A fast reduction in PMN/TL ratio started from day 1 after 5-FU treatment, and a further reduction in PMN/TL ratio occurred from days 7 to 9 PI. On day 15 PI, birds treated with the 5-FU recovered their PMN/TL ratios to the normal. There were highly significant differences (P‹0.001) in PMN/TL ratio between groups, between PI days, as well as in the interaction 5-FU dosage and PI time. A significant reduction(P‹0.05) in the PMN/TL ratio was found in birds given 200 and 300 mg/kg BW 5-FU and between days, compared with the control birds. Clinical signs of toxicity appeared in birds treated with 300 mg/kg body weight 5-FU.
