Motility and fertility of frozen ram semen thawed in two different rhythms of temperature

ABSTRACT
The aim of the study was to compare the effect of two temperatures and thawing rates on motility and fertility of frozen ram semen. A total of 208 ewes and 13 rams of different breeds were used. Semen was collected with an artificial vagina, and it was evaluated, diluted with a Tris-citric acid-fructose-glicerol-yolk extender and centrifugated at 200g/15 min. The sediment was then resuspended with the same extender. Diluted semen was frozen in 0.25 ml straws containing 300 millions of motile sperm. Post-thaw motility was evaluated in 212 straws which were thawed in a water bath at 36°C/8 sec (Group 1) or 70°C/4 sec (Group 2), and another 208 straws were used for intracervical insemination of ewes. Ewes lambed (%) per ewes inseminated with semen thawed at different rates was also evaluated. Post-thaw motility was 51.28 and 47.98% for Groups 1 and 2, respectively, and fertility was 30.7 and 29.2% for the same groups, respectively, without differences between groups (X
2; P›0.05). Thawing straws at 36°C/8 sec is a more practical and safer method.
