In this paper a transpharyngeal technique to hypophysectomize rats is described. The hypopliysis is excised in one piece. A portion of the pars tuberalis always remains attached to the hypothalamus. Pre-and postoperative care is also described. In order to obtain a better survival rate, intermittent aspiration of the mucus through the nostrils and mouth, and the use of an intensive care box (temperature and air controlled) are included. Some criteria to verify the hypophysectomy and some of the most frequent accidents, which can occur during this procedure, are mentioned. The advantages of an adequate operating table, and how to construct some materials and surgical instruments in the laboratory, are described. The average survival rate after hypophysectomy was of 70% in rats from 80 to 130 g bodyweight and of 97% in heavier rats.
