Experimental evaluation of Pasteurella haemolytica immunogen in lambs

ABSTRACT
This study was conducted to evaluate the protective effect of live
Pasteurella haemolytica and its leucotoxin on lambs upon challenge. Forty lambs were used and randomly distributed in four groups. Group one was the control group, receiving subcutaneously only growth media. Group two was subcutaneously immunized with a live culture of
P. haemolytica A1. Group three was intradermically immunized with a live culture of
P. haemolytica. Group four received
P. haemolytica leucotoxin, which was subcutaneously administered with complete Freund's adjuvant. All animals were exposed with PI-3 virus and then challenged with a field strain of
P. haemolytica A1, and subsequently slaughtered to evaluate pulmonary lesions. Control lambs presented fever after the bacterial challenge. Anticapsule and antileukotoxin antibody titrers were higher in all immunized groups compared with the control group (P ‹ 0.05). Pulmonary lesions were more severe in the control group (P ‹ 0:05), but there was no significant difference among the immunized groups. These results show that the protection confered with live
P. haemolytica vaccine or its leucotoxin is satisfactory to protect lambs against pulmonary lesions produced experimentally by the
P. haemolytica challenge. Furthermore, inocuity of the immunogens used in the present study due to the absence of postvacunal reactions, was demonstrated.
