Perfil de expresión de cuatro miRNAs en el suero de pacientes con endometriosis

RESUMEN
Objetivo: Determinar el perfil de expresión del hsa-miR-21, -141, 200b, y -210 en
el suero de pacientes con endometriosis, analizar y establecer las posibles diferencias
entre quienes tienen y no la enfermedad.
Materiales y Métodos: Estudio analítico, longitudinal y prolectivo efectuado en
pacientes atendidas en la Clínica de Infertilidad del Instituto Nacional de Perinatología
Isidro Espinosa de los Reyes de la Ciudad de México entre febrero de 2018 y agosto
de 2019. Criterios de inclusión: pacientes con y sin diagnóstico de endometriosis. A
cada una se le extrajeron 5 mL de sangre para la obtención del suero; a las pacientes
con endometriosis se les reprogramó para electrofulguración y nueva toma de sangre
a la semana. Del suero se extrajo el RNA total y la expresión de los miRNAs se obtuvo
mediante el ensayo de retrotranscripción y PCR. A la banda de expresión se le determinó
la densidad óptica. Para el análisis estadístico se utilizó t de Student; se consideró
significativo el valor de p ‹ 0.05.
Resultados: El hsa-miR-21 y hsa-miR-200b aumentaron significativamente en las
pacientes con endometriosis. Las pacientes que recibieron electrofulguración tuvieron
disminución significativa en la expresión del hsa-miRNA-21 (p = 0.002), -141 (p =
0.03), y -210 (p = 0.009) versus a quienes no se realizó elecrofulguración.
Conclusiones: Las expresiones de hsa-miR-21 y hsa-miR-210b aumentaron significativamente
en las pacientes con endometriosis por lo que estos miRNAs podrían
utilizarse como posibles biomarcadores no invasivos; se han implicado en diversas
vías de señalización inflamatoria, apoptosis y en la diferenciación de la respuesta
inmunológica.
