La Reacción en Cadena de la Polimerasa (PCR) es una de las técnicas mas empleadas en el diagnostico de infecciones por virus; en el caso del Virus de Papiloma Humano no podía ser la excepción. La prueba tiene como fin amplificar regiones génicas de los virus imperceptibles por métodos tradicionales como citología y colposcopía, lo cual permite identificar de manera más exacta la presencia viral y con ayuda de enzimas de digestión (RFLP) realizar cortes exclusivos que hacen a un subtipo viral diferente del otro, para apoyar así a la clasificación del grado oncogénico al cual pertenecen.
