Introduction of a new mixture of solvents in Thin Layer Chromatography

ABSTRACT
Introduction: thin Layer Chromatography (TLC) is one of the choice methods for detecting pharmaceuticals in samples of biological fluids. In it, the mobile phase is a solvent or a mixture of solvents. According to the dissociation constant of each drug, a specific mobile phase is used, for instance, for acid drugs, a mixture of chloroform-ketone is used( 80:20). Chloroform is deemed to be cancerigenic, mutagenic and teratogenic , that is why, is convenient to replace it by other less aggressive solvents.Objective: to introduce a less toxic mixture of solvents cheaper than the one usually employed in the mobile phase of TLC and using half of the volume of the solvents.
Methods: chloroform was substituted by dichloromethane in one occasion and also by dichloroethane , both solvents are less toxic than Chloroform. The Rfs of reference drug patterns supplied by the Cuban Pharmaceutical Industry (Industria Farmacéutica Cubana) were determined in the new solvents systems. The costs for the proposed variants were analyzed too.
Results: new values of Rf were obtained with the introduced solvents, as well as, a save of 17.3 and 40.5 % with the introduction of dichloromethane and dichloroethane respectively.
Conclusions: The introduction of the new solvent mixtures aimed at substituting Chloroform in the TLC, rendered good results and a considerable save of money in foreign currency.
